Change in milliseconds between different fluorescence excitations

With the internal filterwheel it is possible to excite fluorochromes in the range of 20-30ms and switch very fast between three colours and absolutely vibration free.

Using multiple fluorescence labels within the cell is now commonplace for co-localizing events.

  • In the past, this often required expensive external devices like fast-switching monochrometers to switch excitation wavelengths for each probe species
  • Now, the fast IFW in combination with a multi-bandpass emission filter cube allows nearly all GFP variants to be excited within 50 milliseconds

Example:

The BGR filter cube with three excitation ranges 405 to 435, 487 to 502 and 560 to 580.

The internal fast filterwheel consists of a short pass 460 filter, a bandpass 460 to 515 and a long pass 520 and higher. The filters on the wheel preselect a range out of the filtercube.

Furthermore the Leica FIM (Fluorescence Intensity Manager), 2 fast shutters, a motorized Excitation Manager are on a disk with a diameter of 49.5 mm.
